Mami's probably better if you like taking forever (waiting for special to charge), but not really.
On 1st and 2nd levels, Madoka is probably about as fast as Kyoko on round 26 (possibly faster, since her special takes down those big bosses pretty quick). Not the same with 3rd and 4th probably, though Madoka's special really is the best move in the game IMO.
On later rounds, enemies do more damage to you, but you also level a lot quicker. Which means Madoka can often one hit KO, which is quite effective with her strong attack, since it pierces through an entire row of enemies. Kyoko doing more damage doesn't matter as much.

Chances are there's joy pad support.

During the character selection screen, hit the second player's attack key to get his selection to pop up. After you confirmed character selection for both, the game starts and you're playing 2 player.
But you actually need another person with you. Controlling 2 players at the same time is like impossible. One trick is to set player 1 and player 2 to exactly the same keys, and then you can control both that way, but that's not as good as true 2 player and you end up getting hurt more than usual (still much better than 1 player though). If you plan on doing that I suggest you use 2 short ranged or 2 long ranged, not 1 of each.
Will try when I get home, though yeah, don't see much point with only one person.
Could always have long range spamming attacks whilst you control close range character.

Doesn't work because if you press the forward button, both characters would move forwards.
If you want more controlled play, I guess Madoka/Homura works well, sit back and shoot stuff. Your strong attacks together can cover 2 different directions (Madoka's forwards, Homura's goes diagonally up). Also special attack combo is pretty nice, time freeze will allow you to use that arrow thing like 2 times in a row without worrying about getting hit and loosing s
hitloads of HP.
Otherwise just use Kyouko/Sayaka, more erratic gameplay, you just run in and jump around and kekeke everything.
