Squeaky clean 5.00 M33-6 on mah PSP 2000. ISO Tool takes care of 6.XX games. No Prometheus integration here.
I didn't know "5.XX" meant OFW. You didn't specify.
Also, "1.50 Kernal Add-on" ? Kernal ? :D

these are things you don't have not using 620TN-E ;)
-no TN settings,
-no SenseMe,
-no Digital comics, "+" section in XMB
-no New 620Theme's,
-no Password control,
-no game resume(psp-Go), i use this alot,.. can't live without!!
-no isoloader v1.25,
-no HEN cfw, i am kinda starting to prefer HEN,. i def don't like permanent patches,. :/
-no sub folders(some 500 theme's do have sub folders)
-mebbe other some stuff i can't think off,. ;p

You forgot the QWERTY keyboard. But seriously. I don't type on my PSP other than the occasional player name, Google search or nickname. In that order.
- Something like TN settings would be nice. But there's the M33 VSH Menu which pretty much takes care of my CPU speed needs. Can't remember the last time I went to the recovery menu (which is what I think it's trying to emulate) because I activate all my plugins with Notepad.
- I have SensMe - though I don't really use it. At all.

- Comics are usually in .cbz or .cbr format (which are essentially ZIP and RAR archives). PSP Filer opens and zooms them very quickly. I read lots of manga with it -- content which you can't find on PSN.
- Very few themes I like. I'm sure most, if not all of them can be converted to 5.XX using CTFtool GUI.
- Password control ?? Hahaha. I'm an only child. Pørn stash is hidden in the SaveData folder in case someone pokes around.
- "Game resume" sounds nice too but I've been living without it since 2007, so meh. I'll happily continue using the sleep button - like I always have - and turning it off when I'm done (using the vsh menu, much faster than holding the power button up).
- Why would I need an iso loader ? There's the M33 driver. Newer games work. I'm what you would call a happy camper. And the "Speed up MS access" feature from 5.00 M33-6 works wonders. Especially if you have a fake memory stick. You can't find anything similar for 6.XX. Not even fatmsmod, which was the alternative if you were using 5.50 GEN.
- "HEN cfw" isn't even an argument. Homebrew works too but with a lot more compatibility because a lot of homebrew were designed for 3.XX - 5.XX.
- Subfolders for... music ? Yes... because I would really like a really long folder structure like "Music\MP3\Genre\Album\Artist\Tracks". Come on...
One feature from 6.XX that I would love to have would be to set my birthday. Just kidding.

For me HEN is easy no messing with flash0 and works fine,.. i had prefer HEN over permanent patch,. a full custom firmware is not a choice above 6.XX
620TN-E is very fast! if not fastest ;)
Enabling/disabling plugins in TN-E can now be done without a PC,..
There is also some extra feature's added by TN in TN-settings,. ;) its amazing :)
eg unlimited characters in OSK
I have tried some demo/free Comics and the Digital comic reader is way cooler then any homebrew comic reader,. ;p its like reading a digital novel,.. zoom/scroll page flipping etc etc also a intergrated music player,. i am hoping TN will add keys.bin to comics when DCR comes to euro psn,. i currently downloaded the free comics from the US PSN
game resume and birthday is only on the PSP-Go

I have yet to try this "Inferno Driver", but As far as I know it's only to improve compatibility.
Oooh, "unlimited characters in OSK" you say ? And how is that useful exactly ?
I'm curious, how often do you manage your plugins to actually need a different method instead of just using Notepad whenever you want to try a new plugin ? Probably once or twice, maybe if a plugin goes haywire or something (which has never happened to me since I tend to stay away from alpha, beta releases).
Oh, and HEN *does* mess with the flash, it installs to flash0. And when you "cold-boot" it from the memory stick it simply activates the modified components instead of ofw components.

@ DSpider No the Inferno driver is super fast. I have been in the scene since DA (did 3.80M33) sooo the 5.00M33 Speed Up Mem Stick "was" fast, but Inferno is faster.
@Vegetano1 No, ALoader v1.25 does not have the driver. it only has M33 and NP9660. Also no speed up option.
The only thing I don't know is the ME Driver better then the Inferno? I need a user that can go in there flash0, who has ME and grab the MEdriver for me. It should be in the "kd" folder hidden, so you need to show all system files in control panel/folder options in Win XP and 7.
Here is the Inferno driver prx file for people who want to tweak with things. I copied it from my kd folder.

im still in 5.00m33-6 (no promethius), lol
as my ir shell works here i don't want to upgrade unless the irshell updates to latest firmwares

Since you have 5.00M33 could you copy out the OEDriver.prx from the kd folder on flash0 and post it please?
