The thing is, your HDD is unlikely to be able to even read 600MB/sec. The SATA2 cable has a peak transfer rate of 300MB/sec and no HDD actually maxes out the bandwidth. A standard HDD probably can't read faster than 100MB/sec (or if you have a crazy RAID0 setup with 4 SSDs, you could probably get around 300-400MB/sec). Considering other factors, such as fragmentation etc, I'd be surprised if your HDD could read more than 60MB/sec overall for the file.
It does seem the file isn't uploading, but MU rejecting it seems a little unlikely too. How would they know it's a duplicate? Hash the file beforehand? If so, the HDD would still have to read the entire file at least. The only methods it would have could be filesize + filename, though that seems a little problematic...
